Nicolas, in Test Vet, says she was dau of Sir John St John of Bletso. Let's believe this and then address the problem of which of the John St Johns of Bletso it might have been:
From the above her brother was also Sir John and still alive.
The only Sir John this applies to was Sir John St John b. 1495 and d. 1558.
So her father was the John St John who was b. c.1460 and d. 1525.
